Rapeseed -- Determination of chlorophyll content -- spectrometric method
1 Scope of application:
this method adopts spectral method and is suitable for the determination of chlorophyll content in rapeseed. This method cannot be used to determine the chlorophyll content of impact specimen notch projectors in oil, such as impact specimen notch manual broaching machine, impact specimen notch hydraulic broaching machine and impact specimen notch electric broaching machine
2. Principle summary:
extract the sample with a specific extraction solvent in a specific device, and then identify the chlorophyll content in the solution by spectroscopy
3. Main instruments and reagents:
3.1 Instruments
conventional laboratory instruments, mainly 50Hz: analytical balance (accurate to 0.001g), mechanical mill, micro mechanical mill (see Figure 1), filter paper, spectrophotometer (spectral band is widely used in many artificial rubber, resins, including engineering plastics and other resins processed at high temperature, with a width of 2nm), cuvette (1cm), pipette (30ml, ISO class a requirements), test tube with cover
3.2. Reagent
unless otherwise specified, the reagent should be analytical reagent. The main reagent is the extraction solvent [add 100ml absolute ethanol to a 500ml beaker, and then add 300ml anhydrous isooctane (2,2,5-trimethylpentane) or anhydrous industrial n-heptane or anhydrous petroleum ether (basically a hydrocarbon containing 7 carbons, with a boiling point range of 90 ℃ ~ 100 ℃).
